Objective

Proliferation ratio of the 2D/3D cultured cells is quantified by Cell3iMager duos.

Materials and Methods

  • Cell Line: HepG2 cells (RIKEN BRC)
  • Medium: DMEM (Nacalai tesque)
  • Plate
    • 96-well plate flat bottom (Sumitomo Bakelite)
    • 96-well plate U bottom (Sumitomo Bakelite)
    • 96-well plate flat bottom,
    • ultra-low attachment coated (Corning)
  • Seeding cell density: 1000 cells / well

  • Culture days: 4 days
  • Reagent: Hypoxia probe LOX-1(Organogenix)

  • Imaging methods: Bright field, Low-magnification lens

Results and Conclusions

  • This quantification can be alternative method to MTT assay and ATP assay. (Correlation with ATP assay has been confirmed.)
  • It is possible to quantify proliferation ratio of the 2D/3D cultured cells.
  • Incase of 2D cells, proliferation ratio was quantified by Area.
  • Incase of 3D cells, proliferation ratio was quantified by Volume.
