We performed the labeled-free colony formation assay using Cell3iMager duos.
Materials and Methods
Cell Line: HCT116 cells (RIKEN BRC)
Medium: DMEM (Nacalai tesque)
Plate: 6-well plate flat bottom (IWAKI)
Seeding cell density: 50 cells / well
Culture days: 14 days
Imaging methods: Bright field, High magnification lens
Observation: Day2, 7 and 14 (after seeding cells)
Results and Conclusions
Cell3iMager duos can perform the colony formation assay as 'label-free'.
And it is easy to quantify the 'area SUM', 'number' and 'diameter average' of colony.
Since it is a label-free analysis, the time-dependent changes from single cells to colonies can be measure
